What are the possible side effects of antimalarial drugs? Plaquenil is most often well-tolerated but, like with any drug, side effects are possible. Most people about 90% who take antimalarial medications experience no side effects. If common side effects do occur, they are usually minor and last only for a short period of time. The goal with ocular screening for the side effects of Plaquenil is to find the damage at the earliest stage to reduce the amount of vision loss. The risk is low in the beginning and increases over time as the drug collects in the retinal pigment epithelial layer. The risk of damage is dose related. If you are taking hydroxychloroquine for symptoms of rheumatoid arthritis, your symptoms should improve within 6 months. If your rheumatoid arthritis symptoms do not improve, or if they worsen, stop taking the drug and call your doctor. Once you and your doctor are sure the drug works for you.

For prolonged treatment of lupus or arthritis, adverse effects include the acute symptoms, plus altered eye pigmentation, acne, anaemia, bleaching of hair, blisters in mouth and eyes, blood disorders, convulsions, vision difficulties, diminished reflexes, emotional changes, excessive coloring of the skin, hearing loss, hives, itching, liver problems or liver failure, loss of hair, muscle paralysis, weakness or atrophy, nightmares, psoriasis, reading difficulties, tinnitus, skin inflammation and scaling, skin rash, vertigo, weight loss, and occasionally urinary incontinence. The most common adverse effects are a mild nausea and occasional stomach cramps with mild diarrhea. For short-term treatment of acute malaria, adverse effects can include abdominal cramps, diarrhea, heart problems, reduced appetite, headache, nausea and vomiting.
